forskolin on NGM

Hi,

I am intending to grow worms on forskolin treated plates. There are a lot of papers where in they have used a variety of concentration of forskolin on cells. Can anyone help me out by letting me know what the ideal conc of forskolin i need to use in my NGM plates?

I would just try out a dose response curve (steps of 4-10x) to see whatever effect you’re looking for. You often need to add considerably more drug to affect worms than you would cells in culture, and so make sure you make a vehicle only plates (if DMSO, ethanol, or other solvent other than water).

Thanks for that! But what would you think would be an ideal concentration to start with?

This paper used 100µM forskolin (in DMSO, I believe), so that seems like a good point to start with:
