Usually, we leave L4 worms at 30C for 5 hours to generate male worms. Last Friday, I accidently left the worms at 30C for 6.5 hours and found about 10 males in the plate today. However, I wonder if I left worms at 30C for too long, and if it is ok to use the male worms for crossing with other mutants.

The males should be fine.

You might also try heat-shocking for males for one hour at 33C, especially in a waterbath. It works about as well, but it’s faster and you’re less likely to forget your worms and leave them in the incubator.
