I would like to express a tagged version of my protein in a ciliated neuron in order to determine if this protein actually localizes to the cilium.
Any suggestions for a good promoter to use?
I am not sure if it’s better to pick one that expresses in all amphid neurons or to pick one that expresses in a small subset.
Well you could use the promoter of a gene required for ciliagenesis (ie. the BBSome). bbs-5 is expressed exclusively in ciliated neurons, you can look at other bbs genes as well. Hard to say whether it’s better to express in all ciliated neurons or just a subset.
ift-20 expresses well in all the ciliated neurons, as does osm-6. Those might be good to start with!
