I’m doing some genetic crosses with sek-1 and I’m wondering if there’s any reliable phenotype that would be easily identifiable to confirm that it is homozygous. I know there’s the immune function but it seems like it would take to long to test…any thoughts?
Thanks,
Kris
Our lab has been using the sek-1(km4) allele which has a deletion that can be detected by PCR. Using two separate PCR reactions with oligos inside the deletion and outside the deletion allows genotyping of both homozygotes and heterozygotes with confidence. I am happy to share the oligo sequences we use.
Best wishes,
Al
that would be great! the pcr conditions you used would be helpful too.
