I am working on a project which involves half hour time lapse videos of various larval stages and young adults. I have been having trouble finding a slide preparation in which the worms do not sink from the original focal plane in the course of imaging. Does anyone have a protocol for this kind of imaging? Any suggestions of things to try to allow a steady time-lapse?
