I am doing infection experiment using the fungal D.coniospore to infect the L4 stage worms. Surprisingly, I found them become a dense spot of cotton-like hyphae at the third day , but as reported the hyphae shouldn’t appear until at least one week after infection. I wonder whether anybody else have seen such phenomenon? Is it normal to have hyphae appearing so early?

I’m not sure where you got the information that “hyphae shouldn’t appear until at least one week after infection”. In our earliest publication on the subject (Pujol et al. Current Biology, 2001), we wrote, “For the first two days post-infection, worms appear healthy but become increasingly moribund until they die. There is then a very rapid proliferation of the fungus, with extension of multiple hyphae until the worm is hidden by a mass of filaments”. And in Figure 2 in Couillault et al., Nature Immunol, 2004, we described extensive hyphal growth within 3 days: “worms were infected with D. coniospora and when moribund [i.e. 24 h-48 h post-infection] were individually ‘picked’ onto glass slides and placed in a drop of water … Images were obtained after 24 h at 25 °C”.
