Viability Quantification

Can anyone here suggest a method to quantify the viability of C.elegans? I am working with anthelmintic drugs using C.elegans as a model and i am unable to quantify the effect of the drug?

MTT-formazan staining. This and other methods are listed in the forum, which is searchable.

The prob is MTT is not working. The worms seem to die immediately after i add MTT. I figured maybe the prob was with starved worms. So, i tried with well-fed worms, although the worms are alive they do not seem to be forming the blue color.

It’s worked for me and others, so I can’t explain why your experiment is unsuccessful.

Have you searched this forum for alternative methods?

is it an absolute necessity to use starved larvaes for MTT??? what concentrations of MTT do you use???

I followed the protocol from the paper, without modification.

thank u…will try out again with starved larvae so lets see if it works…one more question is 10mg/ml completely soluble in PBS???
