Hi All,
I’m hoping to get suggestions about using protease inhibitor cocktails when preparing worm lysates for western blot.

I’m wondering if the commerically available coctails really block worm proteases effectively, or at least if anyone has used any successfully in doing a western with worm lysate.

In our experience, it will depend on your particular protein. In some examples, at least, “mammalian cell” protease inhibitor mix (with Pepstatin A and Bestatin) worked much better than “general use” protease inhibitor mix. Some few proteins seem to work fine even with no protease inhibitor cocktail at all.
