A third GFP to use with myo-2 and myo-3

I’m looking for a third GFP marker that can be used with and distinguished from myo-2 and myo-3. I tried sur-5, but the signal makes it hard to detect whether myo-2 is there or not. It is for marking a transgene, so I need a DNA construct, not a line with an integrant. I have the Fire Lab kit and am hoping that some other marker in the kit will work. Any suggestions?

Hi Tim,

If you are looking for a bright and reliable marker that is not expressed in muscle, consider unc-122::GFP (unc-122 is also known as ofm-1) which drives expression beautifully in the coelomocytes and nowhere else (in adults).

It’s available at Addgene thanks to Piali Sengupta’s lab, in both rfp and gfp versions; here’s the link to the GFP:

I need a marker that is visible in L4s so I can pick worms during crosses. I’ve seen cc::GFP before but can’t remember if it comes on before the adult stage. Do you remember?

Hi Tim,

unc-122/ofm-1::GFP expression can be seen in embryos. In my impatience, I often check injections 24hrs after finishing and can spot “green eggs.” It is very easily seen in L4s too.
