Any suggestions for RNAis that induce early S phase arrest?

Hi there,

I am looking for RNAis that can induce early S phase arrest or late G1 arrest? Any suggestions would be great!

Though I haven’t used them first hand, some ideas are rnr-1 (knockdown ribonucleotide reductase; nucleotide depletion should lead to stalled forks), orc-1 (or another component of the origin recognition complex; should prevent origin firing), or pcn-1 (essential for DNA replication). RNAi for all three of those is reported to cause embryonic lethality, so good candidates for causing an S-phase arrest.

Alternatively, you could cause an S-phase arrest through use of hydroxyurea, which would be a chemical means to inhibit rnr-1.
