Hi, there,

i want to ask about double knockout C elegans.

Well-known fact rrf-3,eri-1,lin-15 genes silence RNAi function.
Currently,eri-1::lin-15double knockout C elegans already exists in CGC, but rrf-3::eri-1,rrf-3::lin15double knockout C elegans dont exist. Why they dont exist and if these has some reason, please tell me about that.

LIN-15B regulates RNAi, not lin-15A. Don’t use the double mutant, since it confers a very strong Muv and animals grow slowly.

Since you didn’t get answers, I will speculate. At 25C, the rrf-3 and eri-1 single mutants cause reduced brood size and aneuploidy. The double may be sterile at all temperatures. I accidentally found that eri-1 or rrf-3 mutants were sterile in other backgrounds, so this would not surprise me.
