C. elegans RNAi Suppression Help

Hi All,

I’m fairly new to C. elegans, so go easy.

I am an undergrad student and we recently performed an RNAi suppression lab. My professor and I got to talking and started to think of some pretty cool follow up
experiments. He tasked me with finding a way to end with a WT worm through a series of RNAi suppressions. For instance, start with WT > Feed E. coli to mutate to Unc >
Feed E. coli to mutate back to WT. The catch is that the second mutation must control a different gene than the first. Say, some sort of Bli mutation will “fix” the initial Unc
mutation. Is this possible? Does anybody know of an experiment that shows this that can also be done in a simple undergrad setting?

Again, I’m fairly new to working with C. elegans and genetics, but I’m willing to lean. If you have any information, please let me know.
