I would like to use a cell disruptor to break up C.elegans.
Could anyone tell me how I need to prepare the worms for the one-shot adaptor of the constant systems cell disruptor?
I was going to wash them a few times, resuspend 1:1 in lysis buffer, freeze in liquid nitrogen, thaw and use
My worm culture is mixed stage, and my main concern is that the adult worms will block the orifice,
-is this a problem?
- do I need to break the worms up before hand?
I would be most grateful if anyone could share their experience of the prep of worms before loading
