Conditional Over-expression transgenics

Hi,
An undergraduate student wants to overexpress a gene spatiotemporally in different neurons after Day 1 adulthood. Would appreciate if anyone help us in working out the possible strategy here.

I think you’d want to have a two-part system.
First: An inducible Cre or inducible Flippase (like by heatshock).
Second: A plasmid that has your student’s neuronal promoter::lox::STOP::lox::gene or FLP-on equivalent, and inject that as an array.

Other options for a two-part system include tissue-specific rescue of the heat-shock response, split cGal, and Tet/Q.

I would recommend reading Nance and Frokjaer-Jensen’s review in Wormbook, especially the section on bipartite systems.
