There is already a body of work about nicotine exposure in C. elegans. A search of PubMed (https://www.ncbi.nlm.nih.gov/pubmed/) with the term ‘c elegans nicotine’ returns 65 publications, and a number include insulin signaling or TRP channel in the title. You should use that information in the design of your experiment.
