I am looking for a few strains that are expressing mcherry under early embryonic promoters, such as elt-2 or myo-2/-3 or pes-10. I have looked at the CGC but nothing was evident. Please let me know if such strains exist. Thanks
It depends on what you are calling ‘early.’
Early to me means 1-cell to 4-8E (when there are 4-8 gut cells), mid is 8-16E-1.5fold, and late is 2fold-4fold.
myo-2 turns on mid to late (2-3fold). I presume myo-3 would be mid to late.
elt-2 would turn on in 2E stage. That would be early.
If you really want early, you should use something with the pie-1 promoter, which would be maternally deposited and last for most of the early period. There should be TONS of pie-1 driven mCherry strains, such as OD83 (https://cgcdb.msi.umn.edu/strain.php?id=16064).
Good luck!
Steve
Thanks for the reply. To clarify, I am looking for zygotic promoters driving mcherry that turn on before the 100 cell stage. Elt-2 would be perfect, but I am not sure if a elt-2::mcherry line exists. Myo-2::mcherry could work as well.
Alternatively does anyone have the nT1 balancer that contains a mcherry or RFP marker instead of GFP?
THanks!
