FemtoJet Microinjection

Hi, my lab recently purchased the FemtoJet 4x and it requires an external pressure supply. The manual says cleaned nitrogen is an acceptable source, but I was wondering if anyone knew whether that would have any adverse effects on C. elegans. Compressed air that is oil-free, cleaned, and dry is also acceptable and I’m wondering if that would be the better option.

Any help is appreciated!

When you are micro injecting your organism, the pressure is being used to push fluid (e.g., DNA) into the gonad. The air itself should not enter the worm. That being said, nitrogen is inert and should not hurt the worm - air is mostly nitrogen. I have used both sources of pressure without difficulty.

Okay, thank you for the help! Will continue on with using the nitrogen tank.
