I’ve often needed to perform immunofluorescence on a reporter strain to help identify the cells expressing a particular antigen / protein. Or, wanted to increase the signal for a faint reporter. (These days that’s mainly in the ‘satellite’ nematode Pristionchus pacificus.) Typically, the fixation & permeabilization for the antibody staining reduces or eliminates the fluorescence of the reporter - so, one needs an anti-Fluorescent Protein antibody that then works under those conditions. To date, I haven’t found an anti-FP and set of immunostaining conditions that works well.
So, this is a call for anyone that’s been even reasonably successful with an anti-FP to provide information on: 1. Antibody used (catalog no., etc); 2. Staining protocol (or at least general method - e.g., Finney-Ruvkun, Freeze-crack, anti-serotonin protocol, etc.); 3. Worm strain(s) involved, so as to judge the level of expression of the FP, etc.
