Hello everyone,
I have been trying to stain the germline by feeding worms dyes such as Mitotracker and Lysotracker for monitoring mitochondrial and lysosomal health. My experiments are performed in liquid culture in 96-well plates as I’m doing this in conjunction with multiple RNAi knockdowns. However the protocols and concentrations that I find for dye feeding on solid plates do not seem to work in liquid. I have tried multiple dye concentrations between 1-5 UM and the feeding is carried out for ~48 h. I can tell dye uptake is happening from strong intestinal staining and fair amount of staining in other tissues. However the germline doesn’t seem to stain at all. Anyone experienced similar problems with liquid feeding?
