Getting a transgenic line with lower transmissionr ratio for integration

Hi,

I generated transgenic lines expressing extrachromosomal arrays with 80% transmission ratios and soon need to
generate stable lines using UVcross-link integration method. However, my transgenic lines have too high transmission
ratios. I don’t know why. I tried injecting 10ng/ul, 40ng/ul and 50 ng/ul of DNA of interest and always get lines with super
high transmission ratios and can’t use these lines for integration. How can I get a line with a lower transmission ratio
(~30-50%)?

Just do the integration with what you have. You’ll get 2-3 lines instead of 4-5. No big deal.
