Hi guys,
Im looking for a red dye to stain amphids. The mechanism is that my gfp transgene is expressed in one pair of amphids based on the anatomic position, but Im not sure. By staining the amohids with a red dye, I am hoping to see yellow under Fitc.
So far I tried Dil L, the red is too strong and I cannot observe any yellow.
Are there any red dyes with a known amphids stained?

Why don’t you try decreasing the concentration of DiI in your staining protocol? I have used ~1/10th the normal concentration and have gotten very faintly stained amphids that are not too bright to see with GFP.
