A quick question - how many worms does it need to get 1-5 microgram of total RNA? I work on C. remanei and will be doing RNA extraction quite soon. The problem is I need only females so it’s good to know a rough number of worms that are needed I will use RNeasy produced by Qiagen. Thanks a lot!!
Hi Whei-yen,
In our lab we use as a common rule that there are 9ng / adult worm aprox. So we generally use 2000 or 3000 worms per plate: 2000worms = 16ug of total RNA.
Thanks a lot Just got our results back - our goal is at least 1.5 microgram, we tried 100, 200, and 300 worms. It looks like 100 worms is marginally okay but 200 worms is of course on the safe side!
