Large epidermal vesicles?

Hello,

Anyone seen large vesicles like the ones in the image with arrows. These are L1s that have been treated with a compound we are studying. At this dose, the worms arrest with many of these round structures that look like vesicles or multivesicular bodies in the epidermis just under the cuticle. The largest ones are about 1 um with many about 0.5 um in diameter. Are there any good markers for multivesicular bodies in the epidermis?

Thank you,
Keith

Hi Keith,

From a first look these seems like epidermal lipid droplets to me.

Have a look in this paper: Identification of lipid droplet structure-like/resident proteins in Caenorhabditis elegans - ScienceDirect

They come in a wide variety of sizes and can be stained with lipophilic dyes such as OilRed and Bodipy for confirmation or genetic markers such as the ones in the above paper.

Hope that helps.

Cheers,
Igor
