Large scale protein purification protocol

Dear wormers,

I am planning to perform a classical protein purification from worm extracts. This will imply to run chromatography, identify fractions with the enzymatic activity of interest and ultimately send it over for mass-spec identification.
WE need to start from a relatively large worm crude extract and I was wondering if anyone can help me providing a protocol for this purpose.
