looking for unstable fluorescence reporter

Hi,

I am looking for some sort of unstable fluorescence reporter.
I am studying some gene down-regulation and it could be detected by qRT-PCR in 3-hour-treatment of drugs.

However the down-regulation could be detected in a much longer treatment with the promoter::GFP reporter in worms.
I guess one of the possibility is that the current GFP reporter is quite stable and it accumulates.

I am looking for any transient fluorescence reporter that could be expressed in the worms to study gene down-regulation.

Any suggestions will be appreciated.

You can fuse the PEST sequence, which directs proteins to rapid degradataion, from mouse ornithine decarbolxylase to GFP. This has been adapted & shown to work nicely in worms in Frand, Russel & Ruvkun’s paper on molting (PLoS 2005; which you can get by searching PubMed with this #: 16122351). Hope this helps.

I will think about it. I heard some talk today and he mentioned that RFP is more unstable than GFP. Is that true ? Does dsRed behave similarly?
