M9 buffer vs. S basal

Hey,

I have a question concerning flushing worms from plates and washing them. I do infection assays and I always used M9-buffer for everything. I read in a protocol that they are using S basal (which is a base for the liquid medium) instead, does anyone of you know if this is better or less stress for the worms? I’d like to keep the stress as low as possible.

Hi Hellga,

In terms of reducing osmotic stress one could say that M9 (being hypertonic) is not as useful as s-basal (which I believe is isotonic?).

But others will probably give you much better info…it’s just a question of time!
