Does anyone know some reliable methods to measure mRNA turnover in worms? Importantly, not radioactive labeled! Thanks!
Measuring mRNA stability should be possible with 4sU labeling (C. elegans labeling optimized for example in Jungkamp 2011), sampling at several timepoints, then sequencing and analysis approach of how its done in cell lines (enough literature on this). There is no need for radioactivity. If one would like to do pulse chase then I would guess more optimizations are necessary for the pulse and chase conditions. Not sure if anything like this has been done or if there are other c elegans labs already doing it.
