I have recently been trying to perform osmotic avoidance assays using a ring barrier of 4M fructose. The problem is that the positive control I use (osm-9) only escapes at low percentage (40%), while N2 escapes at 15%. I have read that this could be due to dry assay plates, so I have tried playing around with plate conditions but with no further improvement.
I would appreciate all suggestions on the conditions to use for this osmotic avoidance test, e.g. the optimal weight or dryness of the plates.
