Rescue experiment question

Hey

I was wondering if you could help. I want to rescue a double mutant. In particular, I have double mutant X;Y say and I want to see if addition of Y is causing the added phenotype of X. Therefore I want to rescue Y in the double.

I have a wild type worm with Y::GFP translational reporter

How would I go about rescuing Y in the double mutant via crossing?

first I would say you need to (if you have not already done so) read the following intro to worm breeding:

the above link points to just one source of the guide, there are others including the wormbook itself ( http://www.wormbook.org/ ).

BUT…why would you want to do it that way? It’s far easier to rescue your Y mutant using a transgenic construct
(with the the added advantage of a selectable marker that doesn’t make the Worms fat, small, or uncoordinated).

Obviously, it also helps to have access to a transgenics facility…but even if you don’t, you could look for labs nearby that do.

There are thousands of protocols available and several are detailed in the wormbook (so they work).

Genetics is great and sometimes necessary, but here it would not be the first choice.
