We have been performing a great deal of RNAi by feeding, including in some C. elegans strains which are resistant to RNAi. We are about to try to validate RNAi efficacy by qRT-PCR—in other words see if the genes that are supposed to be silenced via RNAi indeed show reduced expression.
I’ve heard people talk of using qRT-PCR to validate RNAi, but I’m surprised to find few reports of it in the literature. Does anyone have any experience with this?
