What is the best method to separte unwanted L1's from an Adult population worms

Was in need of the best advice to know which is the best way to separate unwanted L1s from an adult population. Since, my study would just concentrate on adults, so L1s would great problems later when they change into Adults. So, whats the best way to separate them ? Any suggestions please ??? ??? ???

I don’t know the answer. However, you might want to try glp-4 at 25 C so that they don’t produce progenies.

What is the experiment that you’re trying to perform? If genotype is not critical, then you can use one of the t.s. Spe mutants to prevent the production of progeny. Start with an age-synchronized population by bleach treatment, and propagate those embryos at the restrictive temperature.

If you can’t monkey around with the genotype (or growth temperature), filtering should be a viable alternative. Again, start with an age-synchronized population, and separate on a 20-micron mesh when they reach adulthood and begin to lay progeny. For best results, you’ll need to repeat the filtration every day during the egg-laying period. Start with a large population, b/c you’ll lose a lot of animals along the way. However, samples of 95-98% pure adults are possible by this technique.

If you’re only analyzing a small number of animals, the easiest way is to transfer the adult individuals to fresh plates every day until the reproductive period is complete.
