I have been working on anthelmentic assay. i treat the worms with drug and allow it for recovery overnight in PBS. I do my assay in 1.5ml tubes. the problem is my worms seem to die after overnight incubation. Only about 70% worms survive the next day. Could this be because of oxygen depletion? opening the caps of the tubes also dint solve the problem.
learnt a valuable lesson. Aeration wasnt the problem…i still close the lid of my eppendorf tubes…I was directly using 1X PBS…i prepared 10X PBS diluted it to 1X and my worms seem to show 100% survival…
