Worm soaking in antibodies

Hi Everyone,
Can someone help. I want to soak worms in antibodies with Serotonin. Does any one have any suggestion or protocol.
Thanks.

Mutant2,

I am a bit confused about what you want to do. If you soak worms in any large protein, the cuticle prevents the protein from entering the worms body. Thus, soaking worms in antibody will not result in antibody binding inside the worm. The worms may eat some of the antibody, but that is not generally helpful  On the other hand, small molecules like serotonin can be absorbed through the cuticle - you can either soak the worms in M9 or other liquid containing serotonin or you can put serotonin on plates and let them sit for a few hours before putting on the worms. Note that one generally needs to use high concentrations of serotonin to see an effect on worm behavior, since even serotonin does not flow freely through the cuticle. Please see Anne Harts chapter on Behavior in WormBook for protocols for serotonin exposure (and lots of other good protocols.
